My favourite humour in the show, is when they apply scientific methods to their lives; Operant Conditioning techniques,etc.
Also of course Sheldon's array of expressions, and tics(which I assume to some sort of inexpressible feeling, fighting to get out!). Penny uses a wonderful range of expressions as well.
